• VectorBuilder is an intuitive tool to help your need in developing vectors at an affordable price. I was in need of a CRISPR Lentiviral system with a Puro selection due to working with delicate cells that cannot handle FACS sorting pressures. With their online interface, it was easy to construct the vector I wanted and received the vector in a timely manner! It was simple to produce virus and test it in our cell culture and had good knockdown clones. I definitely will be ordering more vectors from Cyagen to finish up my thesis!

• I have been consistently using VectorBuilder over the last couple years and have been extremely satisfied with the vectors delivered to me. • I have been successfully cloning for 3+ years, but the last time we tried generating KO cell lines with CRISPR/Cas9 vectors, we were not successful. This was upsetting, especially since I invested a lot of time in these efforts. Then, by chance, I attended one of VectorBuilder's seminars. Due to their reasonable prices and the fact I could focus my time on other work, we decided to give their lentiviral (dual gRNA) Cas9 plasmid a try. The turnaround time was reasonable, their customer service was excellent and they actually gave feedback about the status of my cloning projects. Their website is also easy to use. But most importantly: using their Cas9 dual gRNA vector, I was able to successfully generate KO cell lines!

• We applied CRISPR cas9 nickase in our study. VectorBuilder helped us design the gRNAs and construct plasmids with double gRNAs and double selection markers (EGFP/puromycin). The gRNA specificity is quite good, better than available online design tools. We got the knockout cell lines in the first run of electroporation with no off-target events. I would recommend VectorBuilder to CRISPR cas9 users.

Her name is Dr. Swati Roy, and she provided me excellent information regarding their products and vector designs. And then, we recently purchased CRISPR Cas9 dual gRNA vector. Now, we are ordering few more plasmid vectors. As per my experiences, VectorBuilder customer services are excellent and user-friendly. • Before using VectorBuilder, producing lentiviral overexpression vectors for our in vitro models was a recurring and time-consuming enterprise. We faced frequent challenges in every experimental step, which made it difficult to plan the actual experiments. Setting up the process with VectorBuilder was very efficient, thanks to their great expertise and support. Once we started using VectorBuilder's services, we noticed a substantial increase in productivity as well as a more uniform and predictable transduction pattern of our target cells.

  We went on to use VectorBuilder for a large-scale CRISPR screen with a custom library spanning over 1,600 genes. With the provided virus, we were able to easily maintain cell representation and we derived a number of interesting hit genes, which we are excited to follow up on.
